T4 pnk ligase
WebPolynucleotide Kinase. Polynucleotide kinase is used to perform 5’-phosphorylation of DNA and oligonucleotides. When preparing DNA for the ligation step in cloning, either the insert DNA or the vector DNA should contain a 5’ phosphate. Often PCR-amplified DNA is treated with T4 PNK to phosphorylate the 5’ end for subsequent cloning ligation. Web1X T4 DNA Ligase Buffer contains 1 mM ATP and can be substituted in non-radioactive phosphorylations (T4 Polynucleotide Kinase exhibits 100% activity in this buffer). WebWe are excited to announce that all reaction buffers are now BSA-free. NEB began switching our BSA-containing reaction buffers in April 2024 to buffers containing Recombinant Albumin (rAlbumin) for restriction enzymes and some DNA modifying enzymes. Find more details at www.neb.com/BSA-free. WebT4 DNA Ligase is a ligation enzyme that can be used to join DNA fragments by catalyzing the formation of phosphodiester bonds between juxtaposed 5' phosphate and 3' hydroxyl termini in double-stranded DNA using ATP as a coenzyme.

WebCloning help (T4 PNK and T4Ligase) I’ve been working on a cloning scheme for awhile that involves editing an Nterminal HIS tag to a Cterminal by adding a stop codon and a restriction site. I’ve ordered 2, 43bp long oligos (rich in Cs and Gs because Of HiS tag). I’ve annealed them and tried the ligation several times to no avail.
WebJun 3, 2002 · End-joining reactions were carried out as described in (A) using GM00558 extracts, except that XRCC4 antiserum (α-XRCC4, 1:200 dilution) was added where indicated. Following deproteinization, aliquots of DNA (40 ng) were incubated with T4 PNK (5 U) and T4 DNA ligase (80 U) as shown.
